No, because have to change the font feature before you load the font.


When you use \em to get a italic text this is enough:

\definefontfeature [default] [default] [itlc=yes]

\setupbodyfont [pagella,12pt]

\starttext

This {\it will} work?

\stoptext


but to get additional space after italic text when you use the \it 
command you have to add \setupitaliccorecction to your setups:

\definefontfeature [default] [default] [itlc=yes]

\setupbodyfont [pagella,12pt]

\setupitaliccorrection [text]

\starttext

This {\it will} work?

\stoptext
